OmniTaq 3 DNA polymerase is a mutant of Taq polymerase that makes the enzyme resistant to the inhibitory effects of blood, soil, and more.  It remains functional in up to 40% whole blood, especially in the presence of our enhancer products. OmniTaq 3 is suitable for direct amplification of samples containing plant tissues and feces. It also works in some concentrations of crude soil extract or inhibitory food matrices where other commercial enzymes fail.

OmniTaq 3 Tolerates up to 40% Blood in the PCR Reaction

New Taq mutant dramatically outperforms the wild type Taq  and remains functional in at least up to 40% blood.

A 1 kb target was amplified directly from human blood, present at 5-40% in 50 ul reaction, with 0.5 ul “1X” OmniTaq 3 or similar enzyme strength (1 μl/10U) of plain Taq (NEB). No PCR enhancer was used.  Control reactions (C) contained no blood but 10 ng human DNA.                                                             

Results: The new Taq mutant dramatically outperforms the wild type Taq  and remains functional in at least up to 40% blood. This enzyme strength overamplified and smeared the OT3 control.  Up to 5x less enzyme is needed for purified DNA (results not shown).
