We developed our PCR Enhancer Cocktails (PEC) to overcome PCR inhibitors and to permit PCR amplification of GC-rich targets. Combined with our inhibitor-resistant Taq mutants, OmniTaq or Omni Klentaq, PEC enables efficient amplification of exogenous and endogenous targets directly from crude samples containing human plasma, serum, whole blood, soil, feces, and plant tissue without DNA purification. These enhancer cocktails enable the Taq mutants to tolerate at least 25% plasma, serum, or whole blood and as high as 80% GC-content templates. PEC improves real-time PCR performance directly in crude samples with both SYBR Green fluorescence and TaqMan assays. PEC also facilitates DNA amplification from crude samples using most commercial Taq polymerases. (However, PEC does not perform well with AmpliTaq Gold).
GC = PEC for high GC-content targets
Generally, if you are working with PCR samples containing purified DNA, we recommend PEC-1 or, for high GC-content targets, PEC-1-GC. Otherwise, for direct amplification of samples containing known PCR inhibitors we recommend PEC-1 / PEC-1-GC, PEC-2 / PEC-2-GC, or PEC-P, depending on the inhibitor and anticoagulant used and GC content of the target.
